摘要: Abstract Carnosic acid (CA) and carnosol (CS) are two structurally related diterpenes present in rosemary herb (Rosmarinus officinalis). Although several studies have demonstrated that both can scavenge free radicals interfere cellular processes such as cell proliferation, they may not necessarily exert the same effects at molecular level. In this work, a shotgun proteomics study based on stable isotope dimethyl labeling (DML) nano-liquid chromatography-tandem mass spectrometry (nano-LC-MS/MS) has been performed to identify relative changes proteins gain some light specific targets mechanisms of action CA CS HT-29 colon cancer cells. Protein profiles revealed induce different Nrf2-mediated response. Furthermore, examination our data each diterpene affects protein homeostasis by mechanisms. treatment induces expression involved unfolded response concentration dependent manner reflecting ER stress, whereas directly inhibits chymotrypsin-like activity 20S proteasome. conclusion, unbiased proteomics-wide method applied be powerful tool reveal differences bioactive compounds biological model.
