摘要: Abstract Aim We tested the utility of topical application Chloroquine-Solid Lipid Nanoparticle loaded gel against rheumatoid arthritis using Complete Freund's adjuvant (CFA) induced arthritic model. Methods Chloroquine solid lipid nanoparticle (SLN) was prepared probe sonication method. Briefly, drug solubilized into melted compritol which further emulsified aqueous medium followed by subjection to ultra-sonication reduce globule size. Further cooling nano sized emulsion rendered SLN incorporated a matrix. Ex vivo permeability study same also performed Franz diffusion cell, taking chloroquine phosphate as standard. Pharmacodynamics (Radiographic & histopathology analysis) evaluate efficacy formulation in rats. Results with particle size 113.75 nm and entrapment efficiency 97.23% were obtained. It found from ex-vivo permeation that chloroquine-SLN shows maximum retention skin compared gel. Radiographic studies rats treated revealed lesser extent bone cartilage degradation those Conclusion The had greater protective potential over may be possible target affected site locally overcome problems gastrointestinal well dose dependent systemic side effects associated oral administration chloroquine.
