Chromosome-specific DNA Repeat Probes

摘要: In research as well in clinical applications, fluorescence situ hybridization (FISH) has gained increasing popularity a highly sensitive technique to study cytogenetic changes. Today, hundreds of commercially available DNA probes serve the basic needs biomedical community. Widespread however, are often limited by lack appropriately labeled, specific nucleic acid probes. We describe two approaches for an expeditious preparation chromosome-specific DNAs and subsequent probe labeling with reporter molecules choice. The described techniques allow repeat suitable enumeration chromosomes interphase cell nuclei or tissue sections. addition, there is no need chromosome enrichment flow cytometry sorting molecular cloning. Our PCR-based method uses either bacterial artificial human genomic templates {alpha}-satellite-specific primers. Here we demonstrate production fluorochrome-labeled 17 18 just few days without specialized equipment limitation only fluorochrome labels.
