Protein and mRNA content of TcDHH1‐containing mRNPs in Trypanosoma cruzi

摘要: In trypanosomatids, the regulation of gene expression occurs mainly at post-transcriptional level. Previous studies have revealed nontranslated mRNA in Trypanosoma cruzi cytoplasm. Previously, we identified and cloned TcDHH1 protein, a DEAD box RNA helicase. It has been reported that Dhh1 is involved multiple RNA-related processes various eukaryotes. also to accumulate stress granules processing bodies yeast, animal cells, brucei T. cruzi. localized discrete cytoplasmic foci vary depending on life cycle status nutritional conditions. To study composition mRNPs containing TcDHH1, carried out immunoprecipitation assays with anti-TcDHH1 using epimastigote lysates. The protein content was determined by MS pre-immune serum used as control. We ribonomic approach identify mRNAs present within immunoprecipitated complexes. For this purpose, competitive microarray hybridizations were performed against negative controls, nonprecipitated fraction. Our results showed associated stage are those expressed other forms cycle. These data suggest metabolism, regulating least epimastigote-specific genes.
