Effects of insulin-like growth factor receptor inhibition on human melanomas in culture and in athymic mice

摘要: Abstract The role of the insulin-like growth factor (IGF) receptor in regulating melanoma cells was evaluated by examining effect antibody-mediated IGF inhibition on four human cell lines culture and as xenotransplants athymic mice. All expressed typical type I receptors an antibody to this (αIR-3) inhibited [ 125 I]IGF-I binding. However, varied widely their vitro responsiveness IGF-I αIR-3: WM 373 852 lines, stimulated replication αIR-3 response, whereas 239-A 266-4 neither nor affected growth. A wide variation also observed different but qualitatively correlated with responses : treatment significantly did not inhibit or may even have had a slight stimulatory effect. These results indicate that pathway is functional regulator vivo some melanomas reflected activity determined . findings suggest therapies aimed at inhibiting be beneficial treating melanomas.
