摘要: The monocyte is a pivotal cell in septic patients that responds to endotoxin with release of inflammatory cytokines. Monocytes display on their surface receptor (CD14) for complexes formed by (lipopolysaccharide, LPS) and plasma LPS-binding protein (LBP). We compared monocytes obtained from normal controls those expression CD14 flow cytometric analysis immunofluorescent-stained cells. In individuals patients, 75%-95% are positive (CD14+). Mean fluorescence exhibited the CD14+ population was measured after maintaining cells at 37 degrees C 15 minutes baseline held 4 (mean ratio). All increased mean ratio warming; however, level achieved average 78% +/- 8% control levels (p = 0.014). To further clarify expression, we examined effect Escherichia coli LPS comparing treated serum-free buffer (no LBP) whole blood (containing LBP). (1.0 ng/mL) incubated 120 generated an increase buffer. contrast, phorbol myristate acetate lowered levels. These data indicate presence ligand (LBP-LPS complexes) CD14, whereas diminished. conclude were responsive other stimuli aside decreased may poor prognosis.
