Fabrication of aptamer decorated dextran coated nano-graphene oxide for targeted drug delivery.

摘要: In the current study, dextran (DEX) was covalently conjugated to surface of nano-GO sheets, making stable biocompatible coated GO (GO-DEX). The prepared GO-DEX nontoxic 4T1 mammary carcinoma cell line at concentrations up 300μg/mL. AS1411 aptamer, a ssDNA aptamer which can improve intracellular uptake by nucleolin recognition, also has been introduced hydroxyl groups DEX in produce GO-DEX-Apt. Moreover, curcumin (CUR), natural polyphenol, found rhizomes Curcuma longa (turmeric) shows antineoplastic effects, loaded onto and GO-DEX-Apt via π-π stacking interactions with high loading capacity (∼29wt%). GO-DEX-Apt-CUR could efficiently enter into MCF-7 over-expressed cancer cells confirmed fluorescence microscope flowcytometry, it showed significantly higher cytotoxicity. These types targeted nanoscale drug delivery vehicles on basis may find potential application chemotherapy.
