Thymoquinone suppresses metastasis of melanoma cells by inhibition of NLRP3 inflammasome

摘要: The inflammasome is a multi-protein complex which when activated regulates caspase-1 activation and IL-1β IL-18 secretion. NLRP3 (NACHT, LRR, pyrin domain-containing protein 3) constitutively assembled in human melanoma cells. We have examined the inhibitory effect of thymoquinone (2-isopropyl-5-methylbenzo-1,4-quinone), major ingredient black seed obtained from plant Nigella sativa on metastatic (A375) mouse (B16F10) cell lines. assessed whether inhibits metastasis cells by targeting subunit inflammasomes. Using an vitro migration assay, we found that inhibited both was also observed vivo B16F10 model. inhibition accompanied decrease expression resulting proteolytic cleavage caspase-1. Inactivation resulted IL-18. Treatment with NF-κB activity. Furthermore, reactive oxygen species (ROS) partial inactivation inflammasome. Thus, exerts its our results indicate can be potential immunotherapeutic agent not only as adjuvant therapy for melanoma, but also, control prevention melanoma.
