Gene structure and glial expression of the glycine transporter GlyT1 in embryonic and adult rodents

摘要: Na+/Cl(-)-dependent glycine transporters are crucial for the termination of neurotransmission at glycinergic synapses. Two different transporter genes, GlyT1 and GlyT2, have been described. Several isoforms differing in their 5' ends originate from gene. We determined genomic structure murine gene to elucidate genetic basis underlying isoforms. Analysis cDNA 5'-ends revealed that GlyT1a 1b/1c mRNAs transcribed two promoters. During embryonic development were detectable by RNase protection assays as early day E9 reached maximal levels between E13 E15. In situ hybridization expression developing spinal cord mainly ventral part ventricular zone E12. At later stages (E15) transcripts also found lateral half basal intermediate gray matter. contrast, second GlyT2 displayed a completely pattern. E11 it is expressed mantle zone, throughout horns. adult rat brain cord, signals exclusively glial cells. Our data indicate an marker neural encodes glia-specific proteins.
