摘要: Colorectal cancer (CRC) is the third most common in men and second women worldwide. Both genetic epigenetic alterations are CRC driving force of tumorigenesis. The adenoma-carcinoma sequence was proposed 1980s that described transformation normal colorectal epithelium to an adenoma ultimately invasive metastatic tumor. Initial changes start early accumulate as it transforms carcinoma. Chromosomal instability, microsatellite instability CpG island methylator phenotype pathways responsible for cancer. pathway consist activation proto-oncogenes (KRAS) inactivation at least three tumor suppression genes, namely loss APC, p53 heterozogosity (LOH) long arm chromosome 18. Mutations TGFBR PIK3CA genes have also been recently described. Herein we briefly discuss basic concepts integrity consequences defects DNA repair relevant CRC. Epigenetic alterations, essential tumorigenesis, reviewed alongside clinical information
