Novel Acetone Metabolism in a Propane-Utilizing Bacterium, Gordonia sp. Strain TY-5

摘要: In the propane-utilizing bacterium Gordonia sp. strain TY-5, propane was shown to be oxidized 2-propanol and then further acetone. this study, subsequent metabolism of acetone studied. Acetone-induced proteins were found in extracts cells induced by acetone, a gene cluster designated acmAB cloned on basis N-terminal amino acid sequences acetone-induced proteins. The acmA acmB genes encode Baeyer-Villiger monooxygenase (BVMO) esterase, respectively. BVMO encoded purified from TY-5 characterized. exhibited NADPH-dependent oxidation activity for linear ketones (C3 C10) cyclic (C4 C8). Escherichia coli expressing methyl acetate, E. hydrolyzed acetate. Northern blot analyses revealed that polycistronic transcription propane, 2-propanol, These results indicate products play an important role derived clarify pathway (propane → acetate acetic + methanol). This paper provides first evidence BVMO-dependent metabolism.
