Neurons express hemoglobin α- and β-chains in rat and human brains

摘要: Hemoglobin is the oxygen carrier in vertebrate blood erythrocytes. Here we report that hemoglobin chains are expressed mammalian brain neurons and regulated by a mitochondrial toxin. Transcriptome analyses of laser-capture microdissected nigral dopaminergic rats striatal mice revealed presence alpha, adult chain 2 (Hba-a2) beta (Hbb) transcripts, whereas other erythroid markers were not detected. Quantitative reverse transcriptase-polymerase reaction (RT-PCR) analysis confirmed expression Hba-a2 Hbb neurons, gamma-aminobutyric acid (GABA)ergic cortical pyramidal rats. Combined situ hybridization histochemistry immunohistochemistry with neuronal marker nuclear antigen (NeuN) rat further mRNAs neurons. Immunohistochemistry identified alpha- beta-chains both human brains, proteins detected Western blotting whole tissue as well cultures mesencephalic excluding possibility contamination. Systemic administration inhibitor rotenone (2 mg/kg/d, 7d, s.c.) induced marked decrease but neuroglobin or cytoglobin mRNA transcriptome RT-PCR transcriptional downregulation nigral, striatal, Thus, treatments affect mitochondria, opening up they may play novel role function response to injury.
