摘要: A sensitive and reliable method for the determination of total thiamine (vitamin B1) in whole blood has been developed which is suited routine analysis. After extraction, enzymatic hydrolysis phosphate esters, separated from interfering compounds by a fully automated high-performance liquid chromatographic (HPLC) system. Thiamine detected fluorometrically as thiochrome. By calculating concentration on-line with aid computer, it possible to complete one analysis within four hours. Routine determinations can be carried out series 120 samples 48 hours.The within-assay between-assay coefficients variation were 4·2 4·4%, respectively.
