Agonists of the Mas-Related Gene (Mrgs) Orphan Receptors as Novel Mediators of Mast Cell-Sensory Nerve Interactions

摘要: IgE-dependent activation of mast cell is often associated with symptoms attributed to sensory nerves. Depending on the tissues involved such include itching, sneezing, irritation, vasodilation, and reflex secretions. In present study, we hypothesize that neuroactive mediators released from cells may agonists recently discovered orphan receptors referred as nerve specific or products mas related genes. HEK-293 expressing MrgC11 wild-type were loaded calcium indicator Fura-2. A known stimulant RF-amide, neuropeptide FF, evoked a rapid increase in cytosolic but not cells. stimulation either rat basophilic leukemia-2H3 (RBL-2H3 cells) mouse bone marrow-derived cells, substance(s) stimulated increases far exceeded seen control RT-PCR revealed both RBL-2H3 express RF-amide precursor gene proneuropeptide FF (A). Immunohistochemical analysis demonstrated immunoreactivity skin situ isolated skin. These data support hypothesis certain genes participate interactions.
