Photo-protective effect of sargachromenol against UVB radiation-induced damage through modulating cellular antioxidant systems and apoptosis in human keratinocytes

摘要: The aim of this study was to evaluate the photo-preventive effects sargachromenol (SC) against ultraviolet B (UVB)-induced oxidative stress in human keratinocytes via assessing antioxidant properties and underlying molecular mechanisms. SC exhibited a significant scavenging effect on UVB-induced intracellular reactive oxygen species (ROS). attenuated macromolecular damage, including protein carbonyl content, DNA strand break, 8-isoprostane level. Furthermore, decreased Bax, cleaved caspase-9, caspase-3 levels, but increased that Bcl-2, which are well-known key mediators apoptosis. Moreover, superoxide dismutase, catalase, heme oxygenase-1 expression. Pre-treatment with upregulated main transcription factor enzymes, erythroid 2-related 2 level, reduced by UVB irradiation. Extracellular signal-regulated kinase (ERK) Jun N-terminal kinases (JNK) involved regulation many cellular events, treatment reversed ERK JNK activation induced UVB. Collectively, these data indicate can provide remarkable cytoprotection adverse radiation modulating systems, suggest potential developing medical agent for ROS-induced skin diseases.
