Cytotoxic and genotoxic effects of five n-alkanals in primary cultures of rat and human hepatocytes

摘要: Five n-alkanals were examined for cytotoxicity, as evaluated by the trypan blue exclusion test, and genotoxicity, induction of unscheduled DNA synthesis (UDS), in primary cultures rat human hepatocytes. After 20 h exposure, cytotoxicity was similar cells two species, increased with length carbon chain. In hepatocytes, propanal (10-100 mM), butanal pentanal (3-30 mM) hexanal induced a modest but significant dose-dependent increase net nuclear grain counts, while hepatocytes this effect not detected. Nonanal which showed highest cytotoxic effect, failed to induce UDS both cell types. These results seem suggest that at concentrations are presumably attained after ingestion food or generated lipid peroxidation processes five tested unable genotoxic effects liver.
