Frequency and characterization of p53 mutations in primary and metastatic human prostate cancer

摘要: We have studied the frequency of mutations in p53 gene human prostate cancer. The investigated material consisted 20 primary-tumor tissue specimens, obtained by transurethral resection and specimens 15 lymph-node metastases, at total prostatectomy. applied methods encompassed immunohistochemistry on frozen sections, using monoclonal antibody PAb 1801, single-strand conformation polymorphism (SSCP) analysis, after amplification single exon sequences PCR, exons 5 to 8 gene. mutations, leading aberrantly migrating bands PCR-SSCP were identified direct sequencing PCR product. Immunohistochemical analysis completely confirmative. In primary tumors, found 10% (codons 232 273), metastases 15% 248 273). one case (codon same mutation was both tumor metastasis. Our results show that are infrequent metastatic tumors. addition, they indicate there is no strict correlation between
