One-step selection of artificial transcription factors using an in vivo screening system.

摘要: Gene expression is regulated in large part at the level of transcription under control sequence-specific transcriptional regulatory proteins. Therefore, ability to affect gene will using artificial factors would provide researchers with a powerful tool for biotechnology research and drug discovery. Previously, we isolated 56 novel DNA-binding domains from human genome by vivo selection. We hypothesized that these might be more useful regulating higher eukaryotic cells than those selected vitro phage display. However, an unpredictable factor, termed "context effect", associated construction zinc finger factors--- proteins bind specifically 9-base pair target sequences. In this study, directly active protein library. Direct selection constituents library may efficient method isolating multi-finger DNA binding while avoiding context effect.
