摘要: Russell's viper venom (RVV) induced acute renal failure involves both direct and indirect nephrotoxic actions where the specific component/s are yet to be identified. A basic cytotoxin of 7.2kDa (RVV-7) has been identified as potential nephrotoxin. Autoradiographic experiments demonstrated that only RVV-7 among RVV toxins binds specifically mice kidney membrane. Homogeneous preparation confirmed its necrotic cell killing property having EC(50) 4.79+/-3.28microM. Tissue distribution kinetics in showed higher localization compared blood liver. Role inherent factor responsible for was assessed after chemical inactivation cytotoxic activity. Cytotoxicity neutralized by histidine modification but consequent alteration vivo insignificant. Classical concept glomerular capillary wall (GCW) permselectivity barrier denotes apart from size selectivity, GCW also restricts anionic proteins filtration. Reducing pI manipulation surface positive charges resulted decreased accumulation kidney. Histological observations treated with degenerated tubular epithelium.
