A LAMP assay for the rapid and robust assessment of Wolbachia infection in Aedes aegypti under field and laboratory conditions

摘要: With Wolbachia-based arbovirus control programs being scaled and operationalised around the world, cost effective reliable detection of Wolbachia in field samples laboratory stocks is essential for quality control. Here we validate a modified loop-mediated isothermal amplification (LAMP) assay routine scoring mosquitoes from cultures field, applicable to any setting. We show that this rapid robust method highly sensitive specific wAlbB infection within Aedes aegypti under variety conditions. test quantitative nature by evaluating pooled mixtures Wolbachia-infected uninfected it capable estimating frequencies, potentially circumventing need perform large-scale individual analysis status course monitoring. These results indicate LAMP assays are useful screening particularly conditions away facilities.
