Beauvericin alters the expression of genes coding for key proteins of the mitochondrial chain in ovine cumulus-oocyte complexes

摘要: Beauvericin (BEA) is a member of the enniatin family mycotoxins which has received increasing interest because frequent occurrence in food and feed. By its ionophoric properties, BEA able to alter membrane ion permeability uncoupling oxidative phosphorylation. It was also shown oocyte mitochondrial function. In this study, effects at 0.5, 1, ,3 5 μmol/L on expression genes coding for key proteins chain ovine oocytes cumulus cells were evaluated different time points vitro maturation (IVM), germinal vesicle (GV; t = 0), metaphase I (MI; 7 h), II (MII; 24 h). The nuclear (TFAM, NDUFA12, UQCRH, COX4, ATP5O) (ND1, COX1, COX2, ATP6, ATP8) Complexes I, III, IV, V analyzed by qRT-PCR. After exposure, perturbed all observed MI stage (7 h IVM). Expression ND1, COX4 ATP5O downregulated upregulated starting from 0.5 BEA. TFAM, ATP8 1 3 oocytes. Cumulus displayed gene patterns upon exposure. downregulation four complexes could represent major toxic event induced cumulus-oocyte complex may result functional alteration.
