摘要: A fast, simple, and efficient approach, termed consensus multiplex PCRÐRFLP, was developed employed to detect mitochondrial (mt)DNA variation in three orchid species, Spiranthes hongkongensis, S. sinensis, spiralis. Using PCR, pairs of mitchondrial primers were added simultaneously into each reaction tube amplify nonoverlapping introns located the NADH dehydrogenase genes. Fragment length differences PCR amplicons directly detectable between spiralis other two species. Further restriction analysis revealed sufficient mtDNA polymorphism, suitable for phylogenetic studies at interspecific level. This approach is well suited large-scale population surveys genome diversity plants. Additionally, maternal mode inheritance organelle genomes renders this valuable rapid identification origin specific parentage hybrid or allopolyploid
