摘要: Calycosin, the main component extractable from herb Radix astragali, has been shown to inhibit cellular proliferation and induce apoptosis in several cancer cell lines, but underlying mechanisms by way which this occurs remain unclear. In present study, we aimed determine potential effects of calycosin on colorectal cells vitro vivo elucidate molecular action. Colorectal lines SW480 LoVo cervical line HeLa were treated with various concentrations or plus ER beta (ERβ) inhibitor PHTPP. The CCK8 assay, flow cytometry, Hoechst 33258 stain used assess apoptosis. mRNA levels ERβ miR-95 quantified real-time PCR. protein expression ERβ, ERα, IGF-1R, p-Akt evaluated Western blot analysis. We demonstrated that inhibited induced apoptosis, particularly cells, not cells. Calycosin increased reduced alongside down-regulation Inhibiting blocked change resulting increase Additionally, significantly suppressed xenograft tumor growth nude mice. conclusion, exerts an inhibitory effect CRC vitro, through ERβ-mediated regulation PI3K/Akt signaling pathways expression.
