摘要: Genomes of human cancer cells are characterized by numerous chromosomal aberrations uncertain pathogenetic significance. Here, in an inducible mouse model melanoma, we metastatic variants with acquired focal amplification that corresponds to a much larger melanomas. Further analyses identified Nedd9, adaptor protein related p130CAS, as the only gene within minimal common region exhibited amplification-associated overexpression. A series functional, biochemical, and clinical studies established NEDD9 bona fide melanoma metastasis gene. enhanced invasion vitro vivo both normal transformed melanocytes, functionally interacted adhesion kinase modulated contact formation, frequent robust overexpression relative primary melanoma.
