摘要: The aim of this study was the comparison an immunomagnetic separation (IMS)-real-time PCR assay for detection Salmonella with cultural reference method according to §35 German Law on Food and Commodities (LMBG, L 00.00.20:1998). IMS-real-time includes a nonselective preenrichment step, IMS, DNA extraction, as well purification followed by hybridization probe-based real-time analysis. An accurate comparability achieved, because both methods analyzed same preenrichment. evaluation carried out using artificially naturally contaminated meat samples. provides result after 12 13 h. Compared regarding samples, achieved specificity 80% (false-positive rate 20%) sensitivity 100% (false-negative 0%). relative accuracy 94%. limit 10 CFU/25 g. concordance index ?, which defines statistical accordance, 0.85 indicated agreement criteria. analyzing samples (n = 491), showed 99.3% 0.7%) 83.7% 16.3%). 98%. ? had value 0.87 highlighted methods.
