Monoclonal antibodies against human milk-fat globule membranes detecting differentiation antigens of the mammary gland and its tumors

摘要: Mouse monoclonal antibodies have been raised against human milk-fat globule membranes (HMFGM) to obtain reagents for mammary tumor diagnosis. A panel of 17 anti-HMFGM was selected further investigation. Antibody-blocking studies indicated that with these at least nine different non-overlapping epitopes could be distinguished on six molecules, MAM-1 MAM-6. Electron microscopic the cellular localization antigens detected by some revealed they were present cell membrane mainly, microvilli, lining intercellular and intracytoplasmic lumina. The reactivity studied normal tissues in vitro lines. All reacted resting gland while eight also bound breast tumors. None specific or its tumors only, but most other epithelial cells, especially secretory tissues. When tested a variety lines distribution reflecting tissue demonstrated. One nearly all carcinomas their metastases did not react lymphomas, sarcomas, neuroblastomas, melanomas nervous system specificity antibodies, individually, sufficient differential diagnosis carcinomas, when used contribute an important improvement
