摘要: Rabbit hemorrhagic disease virus (RHDV) is the etiological agent of a lethal and contagious rabbits that remains as serious problem worldwide. As this does not replicate in cell culture systems, capsid protein gene has been expressed heterologous hosts or inserted replication-competent viruses order to obtain non-conventional RHDV vaccines. However, due technological safety issues, current vaccines are still prepared from organs infected rabbits. In work, two human type 5 derived replication-defective adenoviruses encoding rabbit VP60 were constructed. The recombinant was multimer mouse lines at levels ranged approximately 120 160 mg/L culture. Mice intravenously subcutaneously inoculated with single 10(8) transfer units (GTU) dose AdVP60 vector (designed for intracellular expression) seroconverted days 7 14 post-immunization, respectively. This generated stronger response than obtained second (AdVP60sec) designed secretion. Rabbits then immunized by parenteral mucosal routes 10(9)GTU antibody evaluated using competition ELISA specific RHDVa. Protective hemagglutination inhibition (HI) titers also promptly detected IgG antibodies corresponding percentages over 85% persisted up one year all rabbits, independently immunization route employed.
