摘要: Abstract Previous treatments of preparative high-performance liquid chromatography (HPLC) have generally assumed Langmuir isotherms, a fixed number surface sites for the sample compounds being separated, and equal molecular weights these compounds. This is consistent with assuming that values column saturation capacity (ws) are various components. However, data summarized in this paper indicate ws can vary widely small molecules. The possibility similar variation larger molecules such as peptides proteins may be even more likely. practical consequences variations solute striking heavily overloaded HPLC separations. separation improve dramatically when second-eluted component has value than first-eluted compound. Conversely, much poorer reversed. When developing procedure under conditions heavy overloading (overlapping bands), it therefore important to know separated.
