摘要: Abstract Craig simulations of preparative high-performance liquid chromatography were carried out for heavily overloaded separations as a function the separation conditions (small-sample retention and column efficiency, sample size). These data used to derive maximum production rate (grams per hour) purified product, results compared with treatment Knox and. Pyper. There is an optimum size plate number every separation; these are related desired recovery product (capacity factor, k ′; α) small under same chromatographic conditions. Relative case 99.8% from feed, 3-fold higher possible if adjusted 95% pure product; 10-fold that give 50% product. Teh required (optimum) halved on going touching-band (99.8% recovery) recovery, further vs . recovery. The also varies ( ′, α sample); value preferred ′ should be between 0.5 3.
