Ascorbic acid protects the diaphragm muscle against myonecrosis in mdx mice

摘要: Objective: Oxidative stress contributes to myonecrosis in the dystrophin-deficient fibers of mdx mice and Duchenne’s muscular dystrophy. We examined effects ascorbic acid (AA), an antioxidant free radical scavenger, on dystrophic diaphragm muscle. Methods: Mdx (14 d old) received AA for 14 d. Control saline. The muscle damage was visualized by penetration Evans blue dye into myofibers extent inflammation assessed histologic analysis. Creatine kinase levels were measured biochemical evaluation fiber degeneration. tumor necrosis factora (a proinflammatory cytokine) 4-hydroxynonenal marker lipid peroxidation) analyzed immunoblotting. Results: Ascorbic decreased creatine levels, myonecrosis, inflammation, factor-a 4-hydroxynonenal. Conclusion: present results suggest that plays a protective role degeneration, possibly decreasing reactive oxygen species, support further investigations as potential therapy dystrophinopathies.
