摘要: The Paf complex of Opisthokonts and plants contains at least five subunits: Paf1, Cdc73, Rtf1, Ctr9, Leo1. Mutations in, or loss subunits have been shown to cause defects in histone modification, mRNA polyadenylation, transcription by RNA polymerase I II. We here investigated trypanosome CTR9, which is essential for survival. results tandem affinity purification suggested that CTR9 associates with homologues Leo1 Cdc73; genes encoding Rtf1 Paf1 were not found. RNAi targeting resulted ten-fold decreases 131 mRNAs: they included several are required gene expression its control, such as those polymerases, exoribonucleases target mRNA, helicases RNA-binding proteins. Simultaneously, some from regions subject chromatin silencing derepressed, possibly a secondary effect the two proteins silencing, ISWI NLP1.
