Induction of Macrophage Differentiation of Human Normal and Leukemic Myeloid Stem Cells by 1,25-Dihydroxyvitamin D3 and Its Fluorinated Analogues

摘要: This study shows for the first time that 1,25-dihydroxyvitamin D3[1,25-(OH)2D3] and two fluorinated analogues of 1,25-(OH)2D3 [24,24-F2-1,25-(OH)2D3 26, 27, 27-F6-1,25-(OH)2D3] induce macrophage differentiation human normal leukemic myeloid stem cells. The addition either or one at concentrations as low 10(-9) M to culture plates containing marrow cells stimulated preferentially differentiate colonies monocytes macrophages. Over 80% were composed only macrophages in 10(-7) analogues. In contrast, control not had less than 35% colonies. Likewise, vitamin D induced colony-forming taken from patients. 10(-8) M, chronic myelogenous leukemia approximately 50% acute nonlymphocytic differentiated macrophage-like about 30 20% cultures patients, respectively. Our findings suggest may play a role hematopoiesis compound related analogue possibly have therapeutic some leukemias.
