摘要: Abstract Chemical profiling of natural products by high performance liquid chromatography (HPLC) was critical for understanding their clinical bioactivities, and sample pretreatment steps have been considered as a bottleneck analysis. Currently, concerted efforts made to develop methods with efficiency, low solvent time consumptions. Here, simple efficient online extraction (OLE) strategy coupled HPLC–diode array detector–quadrupole time-of-flight tandem mass spectrometry (HPLC–DAD–QTOF-MS/MS) developed rapid chemical profiling. For OLE strategy, guard column inserted ground (2 mg) instead loop connected manual injection valve, in which components were directly extracted transferred HPLC–DAD–QTOF-MS/MS system only mobile phase without any extra time, solvent, instrument operation. By comparison offline heat-reflux Citrus paradisi cv. Changshanhuyu (Changshanhuyu) peel, presented higher efficiency perhaps because the pressure gradient elution mode. A total twenty-two secondary metabolites detected according retention times, UV spectra, exact mass, fragmentation ions MS/MS nine them discovered peel first our knowledge. It is concluded that OLE–HPLC–DAD–QTOF-MS/MS offers new perspectives products.
