摘要: RAS genes acquire the most common somatic gain-of-function mutations in human cancer, and almost all of these are located at codons 12, 13, 61, 146. We present a method for detecting K-RAS hotspot 228 cases colorectal cancer. The protocol is based on multiplex amplification exons 2, 3 4 single tube, followed by primer extension PCR products using various sizes primers to detect base changes 61 compared clinicopathological data cancer patients with mutation status. K-RAS occurred 36% (83/228) our cases. Univariate analysis revealed significant association between codon 12 exon 2 poor 5-year survival (p = 0.023) lymph node involvement 0.048). Also, 13 correlates size tumor 0.03). Multivariate adjusted size, histologic grade, metastasis also indicated correlate significantly overall 0.002 0.025). No was observed 146 features. demonstrated simple fast way identify mutation.
