Oct4 Cell-Autonomously Promotes Primitive Endoderm Development in the Mouse Blastocyst

摘要: In embryonic stem (ES) cells and in early mouse embryos, the transcription factor Oct4 is an essential regulator of pluripotency. transcriptional targets have been described ES cell lines; however, molecular mechanisms by which regulates establishment pluripotency epiblast (EPI) not fully elucidated. Here, we show that neither maternal nor zygotic required for formation EPI blastocyst. Rather, first development primitive endoderm (PE), extraembryonic lineage. promote PE fate neighboring secreting Fgf4, expression but promotes cell-autonomously, downstream Fgf4 Mapk. Finally, multiple genes as well metabolic pathways continued growth preimplantation embryo.
