Randomly amplified polymorphic DNA (RAPD) for rapid typing of Lactobacillus plantarum strains

摘要: Randomly amplified polymorphic DNA (RAPD) has been used for rapid typing of Lactobacillus plantarum strains. RAPD was with either purified chromosomal serving as template in the polymerase chain reaction, or crude cell extracts, and using a 9-mer primer 80% G + C content. Amplified visualized by ethidium bromide staining after separation on agarose gels. Patterns from 20 Lact. strains two pentosus were analysed Pearson products moment correlation coefficient (r) unweighted pair group method arithmetic averages (UPGMA). With some exceptions, sources gave same clusters subclusters at similarity level 50%. About 50% could be individually separated all other tested The buffer brand, amount extract PCR-step crucial final pattern.
