Alterations of the erythrocyte membrane proteome and cytoskeleton network during storage - a possible tool to identify autologous blood transfusion

摘要: Mature red blood cells (RBCs) are the end-stage of a development process that starts in bone marrow and continues to differentiate, through reticulocyte stage, entering into circulation with four-month lifespan. While stored, RBCs undergo different changes. The aim this study was evaluate changes occurring RBC membranes during storage could be used as possible markers detect misuse transfusion sports. Whole collected from two volunteers bags stored for 42 days at 4°C. At times (1, 7, 21, storage) whole extracted under sterile conditions submitted membrane ghost preparation further analysis. Proteomic methods were applied using strategies: protein oriented 2-DE gels peptide isobaric tags relative absolute quantitation (iTRAQ). In both approaches, goal compare detectable proteome before after standard times. Some confirmed methodologies employed, while others only one them. Complementarities case showed an advantage. Changes observed complexes. them, consisted proteins decreasing, increasing other RBCs. They mostly located cytoskeleton--spectrin β, band 4.2, ankyrin-1, tropomodulin-1, β adducin, 4.9 (dematin), tropomyosin, some also transmembrane (glycophorin C, aquaporin-1, 3).
