A protocol to analyse cellular dynamics during plant development

摘要: In vivo microscopy generates images that contain complex information on the dynamic behaviour of three-dimensional (3D) objects. As a result, adapted mathematical and computational tools are required to help in their interpretation. Ideally, complete software chain study dynamics 3D object should include: (i) acquisition, (ii) preprocessing (iii) segmentation images, followed by (iv) reconstruction time space (v) final quantitative analysis. Here, we have developed such protocol cell at shoot apical meristem Arabidopsis. The uses serial optical sections made with confocal microscope. It includes specially designed algorithms automate identification lineage analyse surface.
