摘要: Objectives The complete nucleotide sequence of the 12 656 bp plasmid pFPTB1 from Salmonella enterica subsp. serovar Typhimurium, which mediates resistance to tetracyclines and ampicillin, was determined. analysed for potential reading frames structural features indicative transposons transposon relics. Methods Plasmid transformed into Escherichia coli JM109, overlapping restriction fragments were cloned E. vectors sequenced. In vitro susceptibility testing carried out confirm phenotype mediated by this plasmid. Results contains a Tn3-like 4950 consisting left terminal repeat, Tn3-related tnpR tnpA genes transposition functions, novel gene ampicillin bla(TEM-135), right repeat. Immediately downstream, 5215 at end Tn1721-like transposon, including truncated transposase gene, as well tet(A) tetR tetracycline resistance, detected. A 5 direct TAAAA, seen immediately upstream Tn3 part downstream Tn1721 part. also carries replication region similar that Klebsiella pneumoniae pJHCMW1. Conclusion is one few completely sequenced plasmids S. Typhimurium harbours transposon-like structure containing bla(TEM-135) Tn1721-related tetR-tet(A) resistance.
