The Q System: A Repressible Binary System for Transgene Expression, Lineage Tracing, and Mosaic Analysis

摘要: SUMMARY We describe a new repressible binary expression system based on the regulatory genes from Neurospora qa gene cluster. This ‘‘Q system’’ offers attractive features for transgene in Drosophila and mammalian cells: low basal absence of transcriptional activator QF, high QF-induced expression, QF repression by its repressor QS. Additionally, feeding flies quinic acid can relieve QS repression. The Q many applications, including (1) intersectional ‘‘logic gates’’ with GAL4 manipulating patterns, (2) GAL4-independent MARCM analysis, (3) coupled analysis to independently visualize genetically manipulate siblings any cell division. demonstrate utility determining division patterns neuronal lineage function growth proliferation, dissecting neurons responsible olfactory attraction. be expanded other uses organism conducive transgenesis.
