摘要: Three experiments based on an idea of Youderian have been performed to determine the proportions and kinds amino acid substitutions that are detected by gel electrophoresis when applied surveys protein variation in populations. The involved applying sequential method under several conditions pH concentration a large sample human hemoglobins with known substitutions. In first experiment, random 20 different hemoglobin variants was studied, these were separated into 17 distinct electrophoretic classes three conditions, thus giving detectability 85%. A single pass standard eight classes. second experiment compared groups chemically identical, but positions α β chains, while third pairs charge equivalent, at same chain position. distinguished 90% all identical they locations, four out five equivalent site. Examination location each substitution three-dimensional structure showed interior usually less from Hb than surface local interactions spatial neighbors sufficient distinguish very similar outside molecule. "charge ladder" model is clearly incorrect, it appears as practiced our Drosophila has substantial fraction if regarded model.
