Chronic treatment with amlodipine modulates adrenocortical angiotensin II receptors in spontaneously hypertensive rats

摘要: We investigated the effects of long-term treatment with calcium-antagonist, amlodipine, on angiotensin II receptors in adrenal cortex spontaneously hypertensive rats (SHR). Seven-week-old male SHR were treated oral amlodipine (10 mg/kg/day) or vehicle (saline) for four weeks. Age-matched normotensive Wistar-Kyoto (WKY) similar to control SHR. Systolic blood pressure (SBP) showed time-dependent increase but not WKY rats, while significantly reduced high SBP Plasma renin activity was serially increased SHR, which further enhanced by treatment. But plasma aldosterone level changed amlodipine. Competitive reverse transcriptase-polymerase chain reaction that adrenocortical type 1 receptor (AT1R) mRNA progressively decreased vehicle-treated compared and 4-week course AT1R levels comparable those rats. Amlodipine 2 (AT2R) from 8 weeks age. Thus, chronic differently modulates both AT2R a possibly direct manner.
