Protist Diversity and Seasonal Dynamics in Skagerrak Plankton Communities as Revealed by Metabarcoding and Microscopy.

摘要: Protist community composition and seasonal dynamics are of major importance for the production higher trophic levels, such as zooplankton fish. Our aim was to reveal how protist in Skagerrak changes through seasons by combining high-throughput sequencing microscopy plankton collected monthly over two years. The V4 region 18S rRNA gene amplified eukaryote universal primers from total RNA/cDNA. We found a strong variation proportional abundances, difference between depths within euphotic zone. Highest richness late summer-early autumn, lowest winter. Temperature abiotic factor explaining most diversity. Dinoflagellates abundant diverse group followed ciliates diatoms. about 70 new taxa recorded first time Skagerrak. pattern relative read abundance phytoplankton groups well accordance with microscopical biovolumes. This is metabarcoding study all taxonomic Skagerrak, which may serve baseline future surveys effects climate environmental changes.
