Properties of the human nuclear protein p85Mcm. Expression, nuclear localization and interaction with other Mcm proteins.

摘要: Recently we identified a cDNA fragment encoding conserved part of new human minichromosome maintenance (Mcm) protein, provisionally termed P1.1Mcm3. Here, report that the protein is most highly related to yeast cell-division-cycle Cdc47, encoded by open reading frame YBR1441 on chromosome II Saccharomyces cerevisiae. The migrates polyacrylamide gel with an apparent molecular mass 85 kDa and shares areas significant similarity Mcm family replication proteins. It is, therefore, designated as p85Mcm. Microscopic immuno-fluorescence studies revealed p85Mcm located in nuclei interphase cells, but evenly distributed throughout cell during mitosis. amounts do not significantly change cycle, mRNA levels rise beginning S phase. However, vitro differentiation HL60 cells results striking decrease both levels, suggesting role for proliferating, differentiated cells. Under physiological salt conditions, component high molecular-mass complex including other dissociates at ionic strength given stable subcomplexes, one which contains together proteins hCdc21 p105Mcm.
