摘要: In a prelminary communication, we described the establishment of continuous human myeloid cell line (HL-60). Here report detailed properties this and document its derivation from peripheral blood leukocytes patient with acute promyelocytic leukemia. As characterized by light electron microscopy, predominant type in both fresh cultured sources is neutrophilic promyelocyte prominent nuclear/cytoplasmic asynchrony. Up to 10% cells spontaneously differentiate beyond stage, proportion terminally differentiated markedly enhanced compounds known stimulate differentiation mouse (Friend) erythroleukemia cells. The HL-60 lack specific markers for lymphoid cells, but express surface receptors Fc fragment complement (C3), which have been associated granulocytes. They exhibit phagocytic activity responsiveness chemotactic stimulus commensurate mature characteristic transformed form colonies semisolid medium produce subcutaneous tumors (chloromas) nude mice. A source colony-stimulating stimulated cloning efficiency soft agar 5--30-fold. Despite adaptations culture, morphological phenotype chemical induction essentially unchanged through at least 85 passages. Cytogenetic studies reveal aneuploidy. Metaphases 44 chromosomes predominated vivo early culture passages; however, clones 45 or 46 became continued passaging. most consistent karyotypic abnormalities were deletion 5, 8, X addition marker resembling D-group acrocentric submetacentric marker, likely an abnormal E-group chromosome. No DNA herpesvirus RNA retrovirus was isolated provides studying molecular events effects physiologic, pharmacologic, virologic elements on process.
