摘要: Abstract Transverse frozen sections of desheathed rat peripheral nerve were incubated in media different composition prior to fixation and processing for electron microscopic examination. Neurofilaments remained intact when these tissues calcium-free media. A loss neurofilaments their replacement by granular debris occurred myelinated unmyelinated fibers following incubation containing 2 mM calcium. The calcium-mediated disruption was inhibited preincubation or with 1 p-chloromercuribenzoate (PCMB). inhibition PCMB could be partially reversed subsequent 10 dithioerythritol (DTE). Calcium-mediated breakdown did not occur after prolonged media, a finding which suggested that neurofilament dependent upon tissue lost inactivated frozen-sectioned tissues. findings the present study provide morphological evidence mammalian is mediated calcium-activated, PCMB-sensitive enzyme axoplasm fibers.
