摘要: Abstract Aim To evaluate the clinical significance of intratumoral HER2 heterogeneity in gastric cancer (GC). Methods A total 322 GC tissues were evaluated by immunohistochemistry (IHC), which 73 with IHC 2+ or 3+ subjected to fluorescence situ hybridisation (FISH). Also, 3–5 distinct spots each case showing different staining intensities individually comparing intensity gene copy number (GCN). Minimum, average and maximum FISH scores generated for case. Results Intratumoral overexpression amplification 54 30 cases 3+, respectively. These characterised diffuse mixed Lauren type, 2+, low-level amplification. Kaplan–Meier survival analysis revealed that heterogeneous was significantly associated longer disease-free times than homogeneous, high GCN most poor outcome. there a strong correlation between results spot. Quantitative polymerase chain reaction (PCR) cell-free plasma showed quantitative PCR on tissue correlated well those FISH, but level not. Conclusions Considering incidence GC, accurate assessment would require larger more detailed guidelines. The guidelines should include recommendation FISH-scoring areas be selected reference corresponding slide. definition HER2-positive tumours reassessed considering heterogeneity.
