Methods for the Determination of Epithelial Cell Kinetic Parameters of Human Colonic Epithelium Isolated From Surgical and Biopsy Specimens

摘要: Abstract The purpose of this study is to introduce the application new approaches determination human colonic epithelial cell kinetic parameters. isolation pure and intact epithelium from both surgical biopsy specimens forms basis these approaches. isolated used in parameters by (a) flow cytometry, (b) Coulter counting, (c) dried preparations, (d) crypt squashes. Using methods, following results were derived. proportion cells various phases cycle sigmoid was determined be 81.6% ± 2.15% (x SE) G1g0 phase, 15.2% 1.86% S 3.2% 0.62% G 2 + M phases. In rectal epithelium, there 79.6% 3.35% G1G0 16.4% 4.86% 4.08% 1.90% total population ~4.2 × 10 6 5.46 5 per cm , ~2.5 3 1.57 each crypt. Therefore, number crypts square centimeter mucosa could estimated ~1.68 . Lastly, columnar accounted for 76.3% 6.18% cells, whereas remaining 23.7% 4.08%, consisted mucous cells.
