Simultaneous determination of oxytetracycline, tetracycline and chlortetracycline in animal tissues using liquid chromatography, post-column derivatization with aluminium, and fluorescence detection

摘要: A method is described for the simultaneous determination of antibiotics tetracycline, oxytetracycline and chlortetracycline in liver, kidney muscle using liquid chromatography, post-column derivatization with Al3+, fluorescence detection. Samples are extracted by homogenizing glycine–HCl buffer, then applied to cyclohexyl Bond Elut cartridges. These washed water tetracylines eluted methanol. The extracts evaporated dryness under nitrogen residues dissolved an oxalic acid–acetonitrile mobile phase injected onto HPLC system. gradient system used control sample elution. Following elution, tetracyclines derivatized Al3+ ions at 60 °C produce a highly fluorescent derivative. detector set excitation wavelength 390 nm emission 490 nm. assay linear up 2500 ng g–1, has limit 20 230 depending on tissue compound. this laboratory residual concentrations statutory testing schemes.
