摘要: The structure of the gene for human 70-kDa type IV collagenase (gelatinase) was determined. Three overlapping genomic clones were isolated and shown to contain 0.4 kilobase (kb) 5'-flanking region, 27-kb structural gene, 4.5 kb 3'-flanking region. has 13 exons that vary in length from 110 901 base pairs (bp) 12 introns range 175 4350 bp. Alignment intron locations demonstrated 1-4 8-12 coincide with interstitial stromelysin genes, indicating a close relationship these metalloproteinase genes. Exons 5-7 are each 174 bp size, codes one complete internal repeat resembles collagen-binding domains fibronectin. transcription initiation site determined by primer extension S1 nuclease analyses. Analysis 0.4-kb region showed that, contrast genes collagenease stromelysin, there is no TATA box or 12-O-tetradecanoylphorbol-13-acetate-responsive element present promoter whereas two GC boxes. There CAAT box, but potential binding (CCCCAGGC) factor AP-2 located first exon.
